In 2013, it was found to be sold in Japan in synthetic cannabis products. The structure of PB-22 POWDER seems to be based on a knowledge of structure-activity relationships in the indole class of cannabimimetic, but it’s not clear where the idea for its design came from.

QUPIC is a unique chemotype of synthetic cannabinoids because it has an ester linker at the 3-position of the indole, not a ketone like JWH-018 and its similar compounds or an amide like APICA and its similar compounds. For human CB1 receptors, it has an EC50 of 5.1 nM, and for human CB2 receptors, it has an EC50 of 37 nM.

At doses of 0.3–3 mg/kg, PB-22 slows the heart rate and lowers the temperature of rats, which suggests that it has strong cannabinoid-like effects. When given to rats, PB-22 POWDER caused hypothermia that was stronger and lasted longer than JWH-018, AM-2201, UR-144, XLR-11, APICA, or STS-135. Six hours after a dose, the rats’ body temperatures were still lower than they had been before. One clinical toxicology study found that PB-22 POWDER made a person and his dog have seizures.
